Caracterización, inmovilización y aplicación en biocatálisis de la lipasa de Rhizopus oryzae expresada en Pichia pastoris

Abstract

En el presente trabajo se han determinado las propiedades de la lipasa recombinante de Rhizopus oryzae contenida en un extracto proteico obtenido mediante el cultivo de Pichia pastoris (rROL). Posteriormente, se ha estudiado la inmovilización de la lipasa recombinante en soportes de diferente naturaleza, como son el polvo de polipropileno (EP100) y el Eupergit®. Con el objetivo de realizar una comparación, se realizaron los mismos estudios de caracterización e inmovilización con la lipasa nativa contenida en un extracto proteico obtenido del cultivo de Rhizopus oryzae (nROL). Los análisis de caracterización revelaron dos isoformas de la lipasa recombinante con idéntico N-terminal y con masas muy similares. Por otra parte, el extracto nativo mostró tres isoformas de nROL. En cuanto a los estudios de especificidad frente a triacilglicéridos, se determinó un comportamiento igual de nROL y rROL, mientras que con los análisis frente a ésteres de p-nitrofenol se dedujo la presencia de la esterasa en el extracto nativo. En los estudios de inmovilización, tanto nROL como rROL fueron hiperactivadas a ciertas relaciones de actividad ofrecidas por miligramo de soporte. Además, la estabilidad en función de la temperatura y el pH reveló que nROL es más estable que rROL tanto inmovilizada como en solución. Se estudió la síntesis de un aromatizante utilizándose tres derivados inmovilizados: rROL-EP100, rROL-Eupergit®CM y rROL-Sepabeads. Se analizó el efecto de diversas variables sobre la velocidad inicial de reacción y conversión final. El derivado que mostró unos mejores resultados fue el obtenido a partir del EP100 aunque rROL-Sepabeads fue el derivado inmovilizado más estable. Mediante la metodología de la superficie de respuesta, pudo analizarse el efecto de la concentración de ácido butírico y la relación molar butírico:etanol sobre la velocidad inicial de síntesis, la conversión y la producción. Los resultados de velocidad inicial y conversión mostraron un claro efecto inhibidor e inactivación a elevadas concentraciones de butírico. De igual modo un exceso de etanol también resultaba inhibidor. Las condiciones óptimas se determinaron en base a un modelo para maximizar la producción de butirato de etilo. Finalmente, rROL mostró capacidad catalítica en otras reacciones de interés industrial como son, la síntesis de la cortexolona-21-acetato, lípidos estructurados de baja carga calórica además de triacilglicéridos sustitutivos de las grasas presentes en leches maternas.Extracts of mature Rhizopus oryzae lipase overexpressed in Pichia pastoris (rROL) and commercial ones from the native microorganism (nROL) have been characterized. Specific activity of rROL extract was more than 40 fold higher than nROL. The presences of multiple bands of lipases around 34 kDa were both extracts. The influence of ionic strength on optimum temperature and pH was also determined from both extracts, significant differences were observed. Finally, the specificity against triacylglycerol esters and p-nitrophenols esters was also analyzed. Similar behaviour was obtained in front of triacylglycerol esters; however rROL shown an opposite behaviour compared to nROL in front of p-nitrophenol esters with preferences for long chain derivatives because of the presence of an esterase in the commercial product. Two different kinds of support, the polypropylene powder EP100 and Eupergit®C, were tested to immobilize the enzymes. The study of the stability of soluble and immobilized enzymes was performed by means of the Box-Hunter experiment design. The stability of a soluble extracts as well as immobilized derivatives was analyzed and the studies showed higher stability or nROL. In order to apply rROL a flavour synthesis, it was immobilized in three different kinds of support (EP100, Eupergit®CM and Octadecyl-Sepabeads) with the aim of using it in the production of ethyl butyrate. The effect of different parameters on initial reaction rate and yield has been studied for each biocatalyst. The results showed the EP100 derivative as the best choice in terms of initial rate and yield but Sepabeads derivative was the most stable. Moreover, by means of a response surface methodology, the effect of butyric acid concentration and molar ratio butyric acid:ethanol on yield, initial synthesis rate and production was studied. An inhibitory effect as well as inactivation of the enzyme was detected at high concentration of butyric acid. The ethanol also showed an inhibitory effect. The optimum conditions were selected in order to maximise the production. Finally, rROL was used to synthesize cortexolone-21-acetate as well as different kinds of structured lipids, obtaining good result
